Hamas leader Maran Issa was killed in an Israeli airstrike, White House official Jake Sullivan said.
As deputy military commander, Issa would be the most senior Hamas leader to die since the war began on October 7.
The Palestinian group, which controls Gaza, has not officially commented on reports of his death. US Secretary of State Antony Blinken will travel to the Middle East this week on his sixth visit to the region since the outbreak of the conflict in Gaza.
He will hold meetings in Saudi Arabia and Egypt to discuss international efforts to reach a ceasefire agreement that secures the release of all remaining hostages. Negotiations have continued this week in Qatar, despite Israel announcing plans to evacuate more than a million people from the city of Rafah before launching an offensive.
In their first conversation in a month, US President Joe Biden told Israeli Prime Minister Benjamin Netanyahu that this would be a "mistake".
Israeli media sources have reported that Issa was killed in an Israeli airstrike that targeted a tunnel complex under the Nuseirat refugee camp in central Gaza a week ago.
The deputy commander of Hamas' military wing, the Izzedine al-Qassam Brigades, was considered one of Israel's most wanted men. The European Union, which placed the Hamas leader on its terror blacklist, directly linked him to the Oct. 7 attack led by the group that killed roughly 1,200 people and sparked the war.
He was imprisoned by Israel for five years during the First Palestinian Intifada and was detained by the Palestinian Authority in 1997 until the start of the Second Intifada in 2000.
The Israeli military has killed a number of senior Hamas leaders since October 7. Hamas political leader Saleh al-Arouri died in an explosion in the southern Beirut suburb of Dahiyeh. Israel is widely held responsible for that attack.
Sullivan, the White House national security adviser, said other Hamas leaders are believed to be hiding, "likely deep within the Hamas tunnel network" in Gaza.
He pledged that the US would assist the Israelis in the ongoing search for senior Hamas leaders, adding, "justice will come for them as well."
